Purpose To examine the impact of intravitreal aflibercept (IVT-AFL) injection on the 25-item National Eye Institute Visual Function Questionnaire (NEI VFQ) in the COPERNICUS and GALILEO trials. Methods Patients (pts) with macular oedema secondary to central retinal vein occlusion received IVT-AFL 2 mg (IVT-AFL 2q4) or sham monthly for 24 weeks. After Week 24 (W24) in COPERNICUS all pts were eligible to receive IVT-AFL based on visual and anatomical outcomes (IVT-AFL 2q4+PRN, sham+IVT-AFL PRN). A cortical venous infarction model has been evaluated regarding the temporal and spatial evolution of regional flow reduction. Two cortical veins were occluded photochemically with rose bengal and fiberoptic illumination. Seven rats served to demonstrate effects on regional cortical blood flow using laser Doppler scanning. After two-vein occlusion, there was a widespread reduction of flow that gradually deteriorated after vein occlusion when regional cortical flow in a 3.5 x 7.0 mm window: after 15 min it had decreased to 57.8% ± 8.0%, and after 75 min it was 34.3% ± 5.4%. What happens to the ischemic penumbra – defined as a territory of critically reduced blood flow in the close neighborhood of an ischemic core – determines outcome after stroke. Currently the pathophysiology of the penumbra is studied predominantly in rat models with occlusion of the middle cerebral artery. Here we propose two other rat models with distinct advantages. A 41 year old woman found a hard nodule on her external right superior quadrant/axillary prolongation through breast self-examination. Screening of the known candidate genes involved in thyroid organogenesis has revealed mutations in a small subset of patients with congenital hypothyroidism due to thyroid dysgenesis (TD).We studied a girl with TD who had mutations in two transcription factors involved in thyroid development.Sequencing analysis of candidate genes involved in thyroid gland development revealed a new paternally inherited heterozygous mutation in the NKX2.5 gene (S265R) and a new maternally inherited heterozygous mutation in the PAX8 promoter region (-456CT).
